How would one identify the difference between Bacillus anthracus and B. cereus under a light microscope?
I have a bacteria culture and I did a gram stain on it. The bacteria are gram-positive, rod-shaped, chain-forming, facultative anaerobes. These are characteristics shared between both Bacillus anthracis and Bacillus cereus. How would I determine the difference? I don't know if I have a harmless bacteria or a deadly killer in my petri dishes right now. So please help me out with this.
If you're just using a light microscope, then B. cereus generally grows in long chains, whereas B. athracis grows in short chains (2-4 cells) - but in vitro it grows in long chains, so simply looking into a light microscope might not be the best way to differentiate.
If you can grow it, then use sheep blood agar. B. cereus is strongly hemolytic, and it will grow in large, close colonies. B. anthracis is non-hemolytic, and the colonies will be much smaller and discrete. Another option (better for identification) would be to use a selective and differential plating medium, in this case anthracis chromogenic agar (ACA). It isolates and identifies colonies of B. anthracis.
